I am aware of Tezuka's innovations within the industry, but his Disney like character/art style is most certainly worlds apart from today's anime/manga. And there are at least as many anime without big eyes as there are with big eyes (going as far back as Lupin III and a good number of late 70's anime), anime is not inherently a "big eyes" look as so many people ignorantly believe.

While Tezuka is truly a pioneer in the field of manga and anime, many in western Japan cite another manga artist born in almost the same area (Kobe to be exact) that has just as much influence on the modern manga industry today: Mitsuteru Yokoyama. Several of Yokoyama-san's manga series from the 1950's and 1960's, Tetsujin 28-go, Giant Robo, Iga no Kagemaru, and Sally the Witch, all help start various modern genres of manga and anime as we know them today.

Don't forget about the so called curse of Osamu, he did his tv anime for so little an episode others had a hard time. It took Miyazki-San going straight to theaters to break his grip on animation, and He did not make animation financially rewarding, he set the stage for animators working long hours for meager pay hoping for their turn to move up. Tezuka used his personal wealth to do anime at a price per episode others could not match, Astroboy was in high demand so no would could ask for more.
